Abstract

To identify the selected crab species belonging to three families, the present study standardize the DNA-based investigation protocol. The chelate legs are the most reliable body parts for DNA extraction and subsequent experiments related to DNA barcoding. The HCO-LCO primer pairs are efficient at amplifying the targeted mitochondrial COI region. The generated DNA sequences of Portunus pelagicus (family Portunidae), Uca rosea (family Ocypodidae), and Muradium tetragonum (family Sesarmidae) 99-100 % similarity with the reference GenBank sequences by the nucleotide BLAST search. The molecular investigation was congruent with the taxonomic identification of the studied crab species from the Odisha Coast in eastern India
